Background | 胎盤生長因子(Placental growth factor, PIGF/PGF)最初在人類的胎盤中被發(fā)現(xiàn), 預測分子量約為23kDa, 屬于血管內(nèi)皮生長因子(VEGF)家族, PIGF在正常妊娠胎盤及母血中高表達, 在心臟、大腦、甲狀腺和骨路肌肉中少量表達, 男性及正常未孕女性體內(nèi)幾乎檢測不到, PIGF存在4種異構(gòu)體:PIGF-1、PIGF-2、PIGF-3、PIGF-4, 其中PIGF-2對早期(8周)胎盤具有特異性. PIGF通過與受體VEGFR-1/FIt-1特異結(jié)合來實現(xiàn)它的生物學功能. PIGF在胎盤絨毛膜的血管形成中起重要作用, 能促進懷孕早期滋養(yǎng)細胞增殖與分化, 內(nèi)皮細胞增殖和遷移以及抗內(nèi)皮細胞的凋亡; 并能增強VEGF的活性, 促進胎盤多種血管在數(shù)量、分支和大小上持續(xù)增加, 增強血管的通透性, 對胚胎發(fā)育至關(guān)重要. |
